At our lab, we optimized protocols for precise genome engineering and genome-wide functional CRISPR-screening in human cell culture models using different NGS-supported pipelines. Applications of these tools range from studying gene function at the nucleotide level, up to uncovering the full repertoire of human genes that will determine cellular responses to chemicals or therapeutics, for example.
A recently optimized prime-editing protocol allows efficient, allele specific engineering of single point mutations for variant classification, or disease modelling in human cell culture models of choice, possibly including iPS cells.
Fast generation of validated knock-out human cell clones, or isogenic genome-wide screening for gene-gene interaction mapping, are two other examples of our much requested CRISPR applications.
Due to growing demand, we are considering to launch a genome engineering and screening facility for standard and custom-designed applications using CRISPR, combined with a spectrum of functionally distinct Cas proteins.
During our CFG meeting, we aim to highlight the available CRISPR-based possibilities and make an inventory of demands at Amsterdam UMC. We will use these to evaluate the possibilities for facilitary or custom-designed genome engineering and CRISPR-screening services in the near future.
